Single-cell multi-omics (scMulti-omics) offers the ability to quantify multiple modalities simultaneously, revealing the nuanced complexity of cellular mechanisms and their heterogeneity. Inferring active biological networks within diverse cell types, along with their responses to external stimuli, remains a significant challenge for existing tools. Using single-cell multi-omic data, we introduce DeepMAPS, a system for inferring biological networks. A robust learning of relationships between cells and genes, both locally and globally, is achieved by modeling scMulti-omics in a heterogeneous graph using a multi-head graph transformer. DeepMAPS achieved better results in cell clustering and biological network construction than existing tools, as shown by benchmarking. The competitive capacity to derive cell-type-specific biological networks is also evident, utilizing lung tumor leukocyte CITE-seq data, paired with diffuse small lymphocytic lymphoma scRNA-seq and scATAC-seq data. To further enhance the utility and reproducibility of scMulti-omics data analysis, we have deployed a DeepMAPS web server featuring multiple functionalities and insightful visualizations.
Our research project investigated how the level of dietary organic and inorganic iron (Fe) impacted the productive output, egg quality, blood parameters, and iron content in aged hens’ tissues. Sixty-week-old Hy-Line Brown laying hens, totaling 350 birds, were divided into five dietary treatment groups, with each group containing seven replicates. A series of ten cages characterized each replicate. For the basal diet, iron in the form of either organic Fe (Fe-Gly) or inorganic Fe (FeSO4) was supplied, with doses of 100 or 200 mg/kg. A six-week period of ad libitum diet consumption was implemented. Iron supplementation, irrespective of its source (organic or inorganic), led to a statistically significant (p < 0.05) rise in eggshell color intensity and feather iron content, when contrasted with control diets. A statistically important (p<0.005) interaction was noted between iron sources and supplemental dietary levels in relation to egg weight, eggshell strength, and Haugh unit. Organic iron supplementation in the diets of hens led to a statistically significant (p<0.005) increase in eggshell color intensity and hematocrit compared to inorganic iron supplementation. Conclusively, the dietary inclusion of organic iron contributes to a richer eggshell coloration in older laying hens. Four databases, PubMed, Web of Science, Embase, and the Cochrane Library, were searched electronically, with the last date being December 12th, 2022. Meta-analyses facilitated the calculation of mean differences (MDs) or standardized mean differences (SMDs) for blood loss volume (BLV), hematocrit (Hct), hemoglobin (Hb), and operative time, if appropriate.
A qualitative synthesis comprised eleven randomized controlled trials, and a meta-analysis encompassed eight studies. Blood loss volume was reduced by -105 units in the local TXA group compared to the control group (p < 0.000001; 95% confidence interval: -172 to -38). However, the use of local TXA yielded a limited outcome in mitigating the decline in Hct, Hb, and operative duration. A meta-analysis was not feasible because of the differing outcomes in other areas; however, all but one study (showing no significant difference on POD 1) indicated reduced postoperative ecchymosis.
